Well, persistance will build up the calluses, but sometimes during a pretty long gig, my fingertips and hands do get mighty tired and sore (even though I've got some pretty respectable calluses!). To get me through the night I find it is sometimes helpful to take a couple of Tylenol or Advil. They don't zonk you out or anything but they do help.
