Somebody talked about ad-lib lines; I recently heard the line "She'll be drivin' six white horses when she comes" followed each time by "Whoa, Buck!" That just sort of emerged from the audience, from a young lady whose father always did that.
If you have a partner, "When the Saints Go Marching In" can be made to work as a countermelody.
