Seriously, I was delighted to hear back from the local gunsmith that my recently inherited 410 shotgun, and the .22-long Colt revolver, are both in excellent condition, and are ready to go. He is test-firing them, and I will pick them up this afternoon. There is not much to do with them, of course, in our urban environment, but there are a few ugly racoons who keep pawing up my back yard that I would like to meet. Amos, nude bathing is permissible in Yellowstone in the winter -- in fact, you can do it all year 'round. Some of those hot pots will not only remove your clothing, but your skin, body fat, internal organs and all. If you drop your wallet or cell phone in it's best just to let it be.

(I'm not making this up, you know. Pop into some of the hot pots or fumurules and in three or four days you'll be a skim of grease.)

We study ionic current fluctuations in solid-state nanopores over a wide frequency range and present a complete description of the noise characteristics. At low frequencies (f 100 Hz) we observe 1/f-type of noise.

We analyze this low-frequency noise at different salt concentrations and find that the noise power remarkably scales linearly with the inverse number of charge carriers, in agreement with Hooge's relation. We find a Hooge parameter = (1.1 ± 0.1) x 10?4. In the high-frequency regime (f 1 kHz), we can model the increase in current power spectral density with frequency through a calculation of the Johnson noise.

Finally, we use these results to compute the signal-to-noise ratio for DNA translocation for different salt concentrations and nanopore diameters, yielding the parameters for optimal detection efficiency

All cells have evolved strategies to rapidly adapt to shifts in the environment. Cells detect these changes via stimulant molecules that bind to membrane receptors on their surface, which trigger a cascade of intercellular reactions.

However, because these interactions between external stimulants and receptors are so transient, they are extremely tricky to detect and monitor. Gilad Barnea et al. have engineered a highly sensitive assay for identifying these fleeting events. The authors linked three common receptor types?G protein-coupled receptors, receptor tyrosine kinases, and steroid hormone receptors?to a transcription factor via a tether with a cleavage site for a protease.

When these receptors are activated, a protease releases the transcription factor, which then activates a reporter gene in the nucleus. All the genes used in this system originate from outside the cell. These genes thus provide a unique biochemical pathway that is protected from interference from the cell's own signaling molecules. Barnea et al. exploited this assay to identify chemerin as a ligand for the orphan receptor, GPR1. ? B.T.

Next Saturday and Sunday (and possibly Monday) I won't be around much if at all. Pat and I are taking a mid-Winter holiday. We have reservations for a tour of the Firehole River and for a cabin...oh, I forgot to mention...we're going to Yellowstone again. Our cabin will be within a couple hundred yards of Old Faceful and everything. We'll go in from West Yellowstone by snowcoach, tour by snowcoach, and leave by snowcoach on Monday afternoon.

I did a little research in the local newspaper and find that the gas well under our neighborhood was published as operating on Dec. 17. I won't get rich from it, but it would be nice to have a little extra income. I wonder if I have to buy boots and a cowboy hat for a gas lease?

Such exist, and my wife was adamant about my NOT turning the old satellite TV mast in the backyard into one (it's just a piece of pipe that stic vertically from the ground about 6 feet; I don't know how far down it goes but I do know that it's set in a LOT of concrete.

But there are Other Things, and now that I'm well into adulthood I can buy the parts without question. Stuff like LOX and kerosene, or RFNA, or liquid air and alcohol, or...well, let's just say that I'm awaye of the Potentialities.

There are actually videos of young idiots allowing themselves to get shot with one of these things. Bet not many of them are stupid enough to do it again. It's pretty clear there are a lot of kids messing with these things that absolutely do not get how dangerous they are. (And probably some older idiots also.)
